Curcuminoids Modulate Pro-Oxidant Antioxidant Balance but not the Immune response to Heat Shock protein 27 and Oxidized LDL in Obese Individuals

Oxidative stress is involved in pathogenesis of atherosclerosis, because of its involvement in oxidation of LDL cholesterol. Oxidized LDL has well known immunological properties which directly affect the severity and progression of atherosclerosis and also toxic to cells such as endothelial cells. Another consequence of oxidative stress is inducing release of heat shock proteins (Hsps), which protect cells under the stress.
To observe the effect of Curcumin supplementation on overall oxidative stress and circulating levels of anti-oxLDL and anti-Hsp27 in obese individuals.
A 10 week randomized, double-blind, two-arm two period (2x2) cross-over design trial with 30 obese individuals was planned. Eligible patients were randomized to receive either Curcuminoids (1 g/day) or matched placebo for 30 days and then crossed over to the alternate treatment for 30 days following a 2-week washout period. Blood samples were collected at baseline and end of each treatment after 12 h fasting. Anthropometric data were collected before and after each dietary treatment phase.Serum Pro-oxidant antioxidant balance (PAB) along with anti-Hsp27 (anti-Hsp27) and anti-oxLDL titers was measured at the beginning andat the end of each study period.
Results and Discussion:
- There was no significant carry-over effect for any of the assessed parameter between the trial periods in either of the groups.
- Curcumin supplementation was associated with a significant reduction in serum PAB (P=0.044), however, it did not show any significant effect on weight, BMI, anti-Hsp27 and anti-oxLDL values (P>0.05).
This is first cross over trial on Curcumin which showed the effect on pro-oxidant – antioxidant status. Also the result of the study shows that Curcumin mitigates oxidative stress. It is important to note that PAB levels are elevated in patients suffering from Coronary artery disease and acute coronary syndrome, hence PAB reduction by Curcumin in study implies that this phytochemical may have cardioprotective effects.
